The evaluation of bacteriocin cytotoxicity is a critical first step in guaranteeing its safe use in food and therapeutic applications. In this study, the bacteriocins nisin, pediocin, and colicin ([Col] E1, E3, E6, E7, and K) were evaluated for cytotoxicity against cultured mammalian cells. Cellular membrane potential (delta psi) changes of bacteriocin-treated cells were used as an index of cytotoxicity. Simian virus 40-transfected human colon (SV40-HC) cells and African green monkey kidney epithelial cells (Vero) cells were cultured (35°C, 10% CO2, humidified air) as monolayers on tissue culture plates. Log phase cells (~104 cells/mL) were treated with partially purified, individual bacteriocin preparations at 170, 350, and 700 activity units (AU)/mL…

Fujifilm Diosynth Biotechnologies (FDB) is a global contract development and manufacturing organization (CDMO) with over 25 years of experience in process development and/or manufacturing of greater than 310 molecules at sites in: Billingham, England; Research Triangle Park, North Carolina; and College Station, Texas. At our College Station location, we specialize in the development and manufacture of virus-based vaccines (attenuated or recombinant viruses), oncolytic viral therapies (such as adenovirus, polio) and gene therapy vectors (such as adeno-associated virus [AAV])…
The Vero cell line is one of the most widely used continuous cell lines in the world, cited in over 10,000 publications. Though originally developed as a host for viral replication, uses for this highly adaptable cell line have expanded far beyond the research laboratory to include diagnostic practices in hospitals, epidemiological surveys, in vitro fertilization clinics, bacterial toxin assays, and vaccine production. ATCC has played a pivotal role in this expansion by distributing the Vero cell line, ATCC® CCL-81™, to the worldwide scientific research community. Recently, ATCC developed a fully-characterized master cell bank of Vero cells prepared under current good manufacturing practice (cGMP) conditions (ATCC® CCL-81.4™). This report traces the history of the Vero cell line from its origins in the laboratory of Dr. Yosihiro Yasumura to its use as a continuous cell substrate for vaccine manufacturing…
